Solution 1: (NH4)2SO4 1.00 g NaCl 8.20 g MgCl2 x 6 H2O 0.20 g CaCl2 x 2 H2O 0.05 g Trace element solution SL-8 (see medium 745) 1.00 ml Na-DL-malate 3.60 g Yeast extract 1.00 g Na2S2O3 x 5 H2O 0.50 g Distilled water 500.00 ml Solution 2: Potassium phosphate buffer (pH 7.8) 20mM 500.00 ml Boil both solutions under a stream of nitrogen gas for few minutes and autoclave separately under anaerobic conditions at 121?C for 15 minutes. Mix aseptically after autoclaving under a stream of sterile nitrogen gas and adjust final pH to 7.8. Distribute in screw-capped bottles or tubes.
