Peptone 3.00 g
Yeast extract 4.00 g
MgSO4 x 7 H2O 0.10 g
FeSO4 x 7 H2O 18.00 mg
Potassium phosphate buffer (1 M, pH 7.1) 5.00 ml
CaSO4 (saturated aq. solution) 2.50 ml
Sodium resazurin (0.1% w/v) 0.50 ml
NaHCO3 1.00 g
L-Cysteine HCl x H2O 0.30 g
Distilled water 1000.00 ml
Dissolve ingredients (except bicarbonate and cysteine), adjust pH to 7.0, then sparge medium with 100% N2 gas for 30 - 45 min to make it anoxic. Add and dissolve bicarbonate and cysteine, then dispense medium under 100% N2 gas atmosphere into anoxic Hungate-type tubes or seruim vials and autoclave. Adjust pH of complete medium to 7.0 -7.2, if necessary.
