Expression of EBNA-1 permits episomal replication of expression vectors such as pDC406 that contain the EBV origin of replication. The cells can be used as host for expression of transfected DNA.

The line was derived from CV-1 (ATCC CCL-70) by transfection with a plasmid encoding Epstein Barr Virus nuclear antigen 1 (EBNA-1).

Complete Growth Medium

The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.

Subculturing

Protocol: Remove old medium, add fresh 0.25% trypsin solution for 2 to 3 minutes, remove trypsin and let culture stand for 5 to 10 minutes at room temperature. Add fresh medium, aspirate and dispense into new flasks.

Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:6 is recommended

Medium Renewal: Twice per week

Freeze medium: Culture medium, 95%; DMSO, 5%
