Animals were immunized with s1-m1 purified vacuolating cytotoxin (VacA) from Helicobacter pylori strain 60190 (ATCC 49503). VacA is a secreted protein that induces vacuolation of epithelial cells.
Spleen cells were fused with Sp2/0-Ag14 cells.
The antibody reacts with purified native s1-m1 VacA but not with denatured VacA. The antibody recognizes a conformational epitope in the carboxy terminal portion of VacA (amino acids 685 to 821). It does not recognize type s2-m2 VacA.
The antibody can be used for ELISA and immunoprecipitation applications.
