Animals were immunized with a synthetic polypeptide with the sequence LMKLCWKKDPDERPTC corresponding to peptides 502 to 517 of the c-yes oncogene (formerly 770 to 793 of the v-yes oncogene).
Spleen cells were fused with Sp2/0 myeloma cells.
In ELISA tests, the antibody reacts strongly with synthetic peptides representing the yes, abl, and ros oncogenes. Weaker activity against src and fgr was also detected.
In immunoblot assays, a protein of approximately 70 kd is detected in a wide variety of cell lines.
The IgG1 reactivity of the antibody with the v-yes peptide (778 to 793) was confirmed by ELISA. IgG1 was strongly positive while IgG2b was only slightly positive.
